Check what you're buying HA for. If it's skin hydration and fine lines — 120–200mg daily works, with solid meta-analysis backing. If it's knee pain — it helps, but only with mild osteoarthritis and only if your gut bacteria cooperate.
Think of HA as a giant sponge molecule your body makes to keep joints slippery and skin plump. When you take it as a pill, the sponge is too big to squeeze through your gut wall intact — so your gut bacteria have to shred it into tiny pieces first, and those pieces travel to your skin and joints. If your gut bacteria are healthy and diverse, this works surprisingly well. If they're not, the sponge just gets fermented away and nothing reaches its target.
That's the general answer. Your stack is different.

ConditionalCheck your label. If it's for skin: 120–200mg daily and give it 12 weeks. If it's for knee pain: 200–300mg, but only works for mild osteoarthritis — and your gut bacteria have to do most of the job first.
A 2025 meta-analysis of seven trials confirmed the skin result. The knee data is real but conditional — and there's a serious safety flag most brands don't mention (see Safety section before buying).
The Protocol
| Goal | Dose | Timing | Form | When to Assess |
|---|---|---|---|---|
| Skin hydration / wrinkle reduction | 120–200mg/day | With or without food | Full-spectrum or LMW HA | 4 wks (hydration); 12 wks (wrinkles) |
| Mild knee osteoarthritis (OA) | 200–300mg/day | Not specified | LMW or native HMW HA | 2–4 months |
| Older adults (50+) with OA | 200mg/day | Not specified | HMW HA (300kDa–1.5MDa) | 3–6 months |
★ = recommended starting point. No evidence supports a loading phase for any population.
Full-Spectrum HA
Mixed up to 1.8MDa
Best skin wrinkle data (Michelotti 2021). Bio-fermented to mimic the body's own HA range.
Best for: Broad-spectrum skin aging
LMW HA
<50kDa to 500kDa
~16% plasma detection. Less dependent on gut bacteria. Faster acting for skin.
Best for: Skin hydration, mild joint pain
HMW HA
>500kDa to 1.5MDa
~0.2% intact absorption. Needs gut bacteria (Bacteroides) to work. Best joint immune signalling.
Best for: OA via TLR4 signalling
No confirmed food-timing requirement — take with or without meals. HMW HA relies on specific gut bacteria to convert it into bioactive fragments. If you've recently taken antibiotics or eat a low-fibre diet, your gut bacteria may not convert it efficiently. No specific cofactors are required. Avoid concurrent high-dose NSAIDs if trying to assess response — they may mask any benefit.
Safety & Interactions
⚠ Cancer History — Do Not Use
The primary receptor HA binds — called CD44 — is heavily overexpressed on many types of tumour cells. It helps tumour cells migrate and spread. Supplementing with oral HA theoretically provides a substrate that could promote tumour growth or metastasis. This is a mechanistic signal (mostly in vitro), but the severity is high enough that anyone with a cancer history should avoid HA supplements and discuss with their oncologist first.
HA has a mild blood-thinning effect. This compounds significantly if you're taking a combination formula that also contains chondroitin or glucosamine (both have established anticoagulant properties). People taking warfarin, heparin, aspirin, or clopidogrel should have INR monitored and consult their doctor before use.
HA modulates immune signalling through TLR4 receptors. In actively flaring autoimmune conditions, this immune modulation may cause unpredictable effects. Use with caution during disease flares.
No human safety data during pregnancy or lactation. Standard precautionary contraindication applies.
Mild nausea or bloating can occur, particularly with high-dose LMW HA. Resolves on dose reduction or by taking with food.
Upper limit: No formal tolerable upper intake level established by EFSA or NIH. Clinical trials up to 12 months at 400–450mg/day reported no significant adverse events.
Conviction Level
Joint → HIGH: A 12-month, multi-centre RCT (N>500) isolating pure HMW HA (200mg/day) with no combination ingredients and MRI cartilage volume scans alongside WOMAC scores.
Healthy-adult joint → MODERATE: A 16-week RCT in healthy active adults (no OA), measuring inflammatory markers (IL-6, CRP) and joint recovery VAS after exercise, comparing LMW vs HMW HA vs placebo.

How strong is the evidence for the claims in this review? Higher = more confidence the claims are supported. This does not measure how large the effect is or how important it is compared with other levers.
